Israeli Study Finds Viagra May Help Prevent Cancer Spread by Limiting Cholesterol in Tumor Cells
A new Israeli study from the Weizmann Institute of Science suggests that sildenafil, the active ingredient in Viagra, could potentially help prevent the spread of cancer by restricting tumor cells' ability to form metastases. Published in the journal Cancer Research, the research led by Prof. Ayelet Erez and Dr. Yarden Ariav found that sildenafil alters cholesterol transport within cancer cells, reducing the cholesterol available to them. This limits the cells' capacity to detach from the primary tumor and establish new growths in other organs.
The study reveals a previously unknown biological mechanism beyond sildenafil's known effect of dilating blood vessels. Cholesterol, essential for cell membrane integrity, plays a critical role in cancer cell migration and metastasis. By decreasing intracellular cholesterol availability, sildenafil impairs the metabolic changes cancer cells require to spread.
Moreover, the researchers found that combining sildenafil with statins, drugs that lower cholesterol production in the body, could enhance this anti-metastatic effect. While statins reduce cholesterol synthesis, sildenafil limits cholesterol availability inside cells, potentially creating a two-pronged attack on cancer cell metastasis. However, this remains a hypothesis that requires clinical trials to confirm.
To explore real-world relevance, the team analyzed a large database from Clalit Health Services covering about 5 million insured individuals over 20 years. They observed a statistical association between sildenafil use and improved survival among cancer patients, especially those also taking statins. The researchers caution that this observational data does not prove causation, as other factors may influence outcomes.
The findings are based on mouse models, human cancer cell cultures, and epidemiological data, and do not yet constitute proof that Viagra can treat cancer in humans. Prof. Erez emphasized that cancer biology is influenced not only by tumor mutations but also by the patient's metabolic state and concurrent medications. If future clinical studies confirm these results, Viagra could gain a novel role in combating cancer metastasis, a leading cause of cancer mortality.
