The U.S. Food and Drug Administration (FDA) unexpectedly approved Rasonque, a new drug for metastatic pancreatic cancer, developed by Revolution Medicines. The company's market value surged to $46 billion after its stock jumped 477% in the past year, driven by positive news about the drug. Rasonque was approved based on a clinical trial involving about 500 patients who had previously failed other treatments. The drug extended patients' median survival from 6.7 months with chemotherapy to 13.2 months, with fewer severe side effects and improved quality of life. The FDA described the results as unprecedented for this cancer type and granted approval six and a half months ahead of schedule. The trial results were applauded at the ASCO cancer conference in May, marking the first time metastatic pancreatic cancer patients lived over a year after treatment initiation. Revolution Medicines is also testing Rasonque as a first-line treatment, potentially extending survival further.

Rasonque targets the RAS protein, a key driver in pancreatic cancer cells, which accounts for 90%-95% of new pancreatic cancer cases and about 3.2% of all cancer diagnoses. The drug works uniquely by binding RAS to another protein that inhibits its cancer-promoting activity. Revolution Medicines has additional drugs in development targeting RAS in other cancers, including lung cancer. The company was founded by Iranian-American researcher Prof. Kuan Shokat and acquired Warp Drive Bio, which developed the protein-binding technology, in 2018.

Analysts estimate Rasonque’s peak market potential at $11.8 billion if approved for earlier-stage patients. The company’s stock also reflects expectations for its other pipeline drugs and possible acquisition by a major pharmaceutical firm. Merck attempted to acquire Revolution Medicines for $28-$32 billion in January but the deal fell through due to pricing disagreements. Other pharmaceutical giants like Eli Lilly and Pfizer are developing RAS-targeting drugs, but none have matched Rasonque’s clinical success in pancreatic cancer. Israeli companies Silexion and Purple Biotech are also working on KRAS-related therapies, though with smaller market valuations.

This approval marks a significant breakthrough in pancreatic cancer treatment, offering new hope for patients with this aggressive disease and potentially expanding to other cancer types in the future.
