A new drug combination, tarlatamab with atezolizumab, has reduced the risk of death by 58% in patients with small cell lung cancer. The Phase 3 trial showed a median survival of 21.1 months compared to 12.7 months with atezolizumab alone. Presented at the ESMO congress, the results were met with significant enthusiasm. The combination therapy, while causing more side effects, demonstrated substantial improvements in survival and disease control.

  • A new drug combination reduced death risk by 58% in small cell lung cancer patients.
  • Median survival extended to 21.1 months with tarlatamab and atezolizumab combination.
  • The combination therapy showed a 58% reduction in mortality risk.
  • Results were presented at the European Society for Medical Oncology (ESMO) congress.
  • Professor Ido Wolff called the results "amazing" and "even more exciting" than pancreatic cancer breakthroughs.
  • The treatment involves a bispecific antibody targeting cancer cells and immune cells.

A novel drug combination has shown a significant 58% reduction in the risk of death for patients with small cell lung cancer (SCLC), an aggressive and difficult-to-treat form of the disease. The findings, published in the New England Journal of Medicine and presented at the European Society for Medical Oncology (ESMO) congress in Madrid, have generated considerable excitement within the medical community.

In a Phase 3 trial involving 563 patients whose disease had not progressed after initial chemotherapy and immunotherapy, the combination of tarlatamab (Imdelltra) from Amgen and atezolizumab (Imfinzi) from AstraZeneca was compared to atezolizumab alone as maintenance therapy. The study revealed that the tarlatamab combination extended the median survival to 21.1 months, compared to 12.7 months for atezolizumab alone, a difference of over eight months. Furthermore, 79.4% of patients in the combination group were alive after one year, versus 53.3% in the control group.

Professor Ido Wolff, Director of the Oncology Division at Tel Aviv Medical Center, described the results as "amazing" and "even more exciting" than recent breakthroughs in pancreatic cancer treatment, noting that the combination therapy extended median survival for SCLC patients to over 21 months, a "crazy number" for this disease.

Tarlatamab is a bispecific antibody that targets the DLL3 protein on cancer cells and CD3 on T-cells, bringing immune cells closer to the tumor to destroy it. While the combination therapy led to more severe side effects, including cytokine release syndrome and neurological issues, only 5.7% of patients discontinued tarlatamab due to these adverse events. The median duration of treatment was also significantly longer in the combination arm.

Researchers highlighted that the benefits were observed across various patient subgroups, including those with brain or liver metastases, who typically have a poorer prognosis. Dr. Jacob Sands of the Dana-Farber Cancer Institute noted the "remarkable" benefit and consistency of tarlatamab in this patient population. Amgen plans to submit these findings to global regulatory authorities, and the drug is also being investigated in earlier stages of treatment and disease.
